Rapid establishment of EHEC detection method in China

Recently, bacteriological scientists and biotechnologists in China have joined forces to rapidly establish a detection method and identification method for enterohaemorrhagic E. coli O104H4 and have begun to promote it throughout the country. Dr. Xu Jianguo, director of the Infectious Diseases Prevention and Control Institute of China CDC (Center for Disease Control and Prevention), said in an exclusive interview with this reporter recently that the establishment of this method has provided a powerful weapon for the timely detection of this newly emerging pathogen that may invade the country.

Xu Jianguo introduced that there are three common EHEC serotypes: O157, O26, and O111; there are more than 40 uncommon serotypes. O104H4, an outbreak that caused an outbreak in Germany recently, is a rare serotype in the EHEC family. It contains the gene for Shiga toxin 2 (vtx2a) and the three genes on the intestinal accumulation-associated E. coli virulence plasmid: aatA, aggR, Aap.

The CDC Institute for the Prevention and Control of Infectious Diseases in China has previously established a detection and identification method for EHEC-O157H7, and has also established a detection method for virulence plasmids with intestinal accumulating adhesion to E. coli. As a result, the Institute has rapidly established four genes (Shiga toxin 1, 2, hemolysin, EAE islands) for EHEC, and aatA, aggR, and aap on virulence plasmids for intestinal accumulation-associated E. coli. Three genes, as well as detection methods for the O104 and H4 genes. The team of Professor Wang Lei from TEDA Biotechnology College of Nankai University cooperated with the China CDC Institute for the Prevention and Control of Infectious Diseases to establish diagnostic sera covering nearly 50 EHECs and diagnostic sera covering more than 10 species of E. coli H (flagellum) antigens, including O104 and H4. At the same time, a PCR detection method and diagnostic serum capable of detecting O104H4 were established.

It is reported that the detection methods established by the two scientists' teams can not only discover EHEC-O104H4 in time, but also detect other serotypes of EHEC in time.
